چکیده
هدف: هوا به عنوان منبعی از ریزاندامگان ( میکروارگانیسمها ) دارای نقش مهمی در ایجاد آلودگیهای بیولوژیک و آسیبهای جدی در منابع آرشیوی میباشد؛ از اینرو پایش میکروبی منظم هوای مخازن (در راستای سیاستهای حفاظت پیشگیرانه) برای شناسایی و اصلاح شرایط بحرانی، اجتناب ناپذیر است. هدف از این مطالعه ارزیابی سطوح آلودگی بیولوژیک هوا در مخازن نگهداری از منابع آرشیوی سازمان اسناد و کتابخانه ملی ج.ا.ا، با روشهای نمونهبرداری فعال و غیر فعال، و بررسی وجود ارتباط بین نتایج حاصل از این دو روش میباشد.
روشها:در این مطالعه نمونهبرداری از هوای مخازن به دو روش: فعال (استفاده از دستگاه ایرسمپلر)، و غیر فعال (روش پلیتگذاری) بر اساس استاندارد ISO 14698-1:2003، انجام شد. پس ازتعیین تعداد کلی ریزاندامگان زنده و قابل کشت (TVC)، و شناسایی آنها با روشهای میکروبیولوژی، همبستگی میان نتایج حاصل از دو روش و توزیع فراوانی گروههای مختلف مورد ارزیابی قرار گرفت.
یافتهها: در میان قارچهای جدا شده جنسهای Penicillium، Cladosporium و Aspergillus دارای بالاترین فراوانی در هردو روش نمونهگیری فعال و غیر فعال بودند. بررسی درصد فراوانی باکتریهای آلاینده هوای مخازن، در هر دو روش نمونهبرداری، نشاندهنده غالبیت باکتریهای گرم مثبت میلهای ( و رشتهای) اسپوردار، از قبیل گونههای جنسBacillusو Streptomyces، میباشد. تحلیل آماری با نرم افزار SPSS ، همبستگی دو روش ارزیابی کننده کیفیت هوا را تائید نمود. مقایسه توزیع فراوانی گروههای مختلف نشانگر عملکرد بهتر روش فعال برای جداسازی جنسهای قارچی میباشد.
نتیجهگیری: نتایج حاصل از هر دو روش به منظور ارزیابی کلی آلودگی هوا، درپایش های ادواری مخازن، قابل استناد میباشد.

A comparison of Active and Passive air sampling methods in the repositories of archival materials

Purpose: Air as a source of microorganisms has an important role in the biological contamination and serious bio-deterioration of archival materials, so regular microbial monitoring of the air of repositories along with protective conservation policies for identifying and correcting critical conditions, is inevitable .The aim of this study was to evaluate the biological air contamination levels in the archival repositories of the National Library and Archives of Iran, by active and passive sampling methods, and to assess the relationship between the results of the two methods.
Method/Research Design: Air sampling of repositories was carried out in two ways: Active (using Air sampler model, SAS), and Passive (Settle plates) methods, according ISO 14698-1: 2003, standard. After determining of TVC (Total Viable Count) and identifying different groups of microorganisms by microbiological methods, the correlation between the results of the two methods and frequency of different groups were evaluated.
Findings:Among the isolates of fungal genera thehighest frequency is reached byPenicillium, Cladasporium and Aspergillus in both passive and active sampling methods. Prevalence of contaminantbacteria of the air of repositories, in both sampling methods, indicates dominance of sporulated gram-positive rod bacteria such asBacillusandStreptomyces. Statistical analysis using SPSS confirmed thecorrelation between the two methods of air quality assessment. Frequency of different groups of microorganismsintroduces active air sampling as a better method for isolation of fungi genera.
Conclusion: The results of both methods are reliable for periodical monitoring of air microbial contamination.
